Kombucha Tea as a Reservoir of Cellulose Producing Bacteria: Assessing Diversity among Komagataeibacter Isolates

چکیده

Bacterial cellulose (BC) is receiving a great deal of attention due to its unique properties such as high purity, water retention capacity, mechanical strength, and biocompatibility. However, the production BC has been limited because associated costs low productivity. In light this, isolation new producing bacteria selection highly productive strains become prominent issue. Kombucha tea fermented beverage in which fraction microbial community composed mostly belonging genus Komagataeibacter. this study, trials were performed starting from previous batch, bacterial was conducted along cultivation time. From whole pool, 46 isolates tested for their ability produce BC. The obtained yield ranged 0.59 g/L, isolate K2G36, 23 g/L K2G30—which used reference strain. genetic intraspecific diversity investigated using two repetitive-sequence-based PCR typing methods: enterobacterial repetitive intergenic consensus (ERIC) elements (GTG)5 sequences, respectively. results different approaches revealed suitability fingerprint techniques, showing discrimination power, calculated D index, 0.94 rep-PCR 0.95 ERIC rep-PCR. order improve sensitivity applied method, combined model genotyping experiments performed, allowing discriminate among strains.

Nitrogen-fixing and cellulose-producing Gluconacetobacter kombuchae sp. nov., isolated from Kombucha tea.

A few members of the family Acetobacteraceae are cellulose-producers, while only six members fix nitrogen. Bacterial strain RG3T, isolated from Kombucha tea, displays both of these characteristics. A high bootstrap value in the 16S rRNA gene sequence-based phylogenetic analysis supported the position of this strain within the genus Gluconacetobacter, with Gluconacetobacter hansenii LMG 1527T as...
